Abstract:The genetic diversity was analyzed in the back muscles of wild populations of mantis shrimp Oratosqilla oratoria sampled in the sea areas of Qinhuangdao, Tangshan, Changli, Tianjin and Huanghua by a single layer gill net in November 2019 by 12 pairs of microsatellite primers. The results showed that the five populations had from 3 to 16 in the allele number with the average effective alleles ranging from 2.9063 to 3.4102, the average observed heterozgosity (Ho) ranging from 0.5004 to 0.5342, average expected heterozgosity (He) from 0.5310 to 0.5491, and average polymorphic information content (PIC) ranging from 0.5422 to 0.6038. Hardy-Weinberger equilibrium test revealed that 81.67% position was in balance in the five populations. There was the maximal genetic distance between Tianjin and Tangshan populations, and the minimal genetic identity between Tianjin and Tangshan populations. The UPGMA phylogentic tree showed that the populations of Qinhuangdao and Tangshan were clustered together, Changli and Huanghua were clustered together, and then they got together with the Tianjin population. The findings indicated that the five mantis shrimp populations had high levels of genetic diversity, being the potential as breeding material and used as breeding material.
