Molecular Identification of Two Snailfish,Liparis tanakae and L .zonatus in the Liaoning Coast based on COⅠ Gene Sequences |

Abstract Liparis tanakae and Liparis zonatus are two common snailfishes in the Bohai Sea and the Yellow Sea .A total of 20 individuals (9 L .tanakae individuals and 11 L .zonatus individuals) collected from Liaodong Bay were analyzed by sequence variations of mitochondrial CO Ⅰ gene fragments in this study . PCR amplification products of 620 bp COⅠ gene fragments were obtained ,with average contents of 22 .4%in A ,32 .2% in T ,26 .9% in C and 18 .5% in G .Sequence comparison of this segment revealed only two variant sites in the two species (one in L .tanakae individuals and two in L .zonatus individuals) with 2 transitions ,and no transversions and indels .The results showed that genetic distance of intra‐species of L . tanakae was ranged from 0 to 0 .2% and L .zonatus 0 to 0 .3% ,and the genetic distance among species was 0-0 .2% .The mean intra‐species distance was 0 .001 ,and the mean inter‐species distance between the two species was 0 .001 .Integrating morphological and mitochondrial DNA data ,L .tanakae and L .zonatus are considered as the same species of different morphs .
