Abstract Sex steroid hormones play an important role in sex differentiation, reproduction and development of animals. However, it is still unclear whether there are the synthesis, transformation, and transportation of sex steroid hormones in mollusks. In this study, we used enzyme-linked immunosorbent assay (ELISA) to analyze the distribution of progesterone, testosterone and estradiol in tissues of adult female and male Yesso scallop Patinopecten yessoensis. Results showed that progesterone accounted for about 80% of total hormone content in hepatopancreas, significantly higher than that in the other tissues, suggesting that progesterone may be synthesized or absorbed in hepatopancreas; the maximal level of steroid hormones was observed in the hemolymph, suggesting that steroid hormones may be accumulated in hemolymph; the levels of the three hormones were significantly positively correlated, indicating potential conversion of the three hormones in the scallop. No much difference in the hormone levels was found between male and female scallops in most tissues, but estradiol/testosterone was significantly higher in females than that in the gonad and hepatopancreas in males, indicating that the ability to produce estradiol was higher in the female gonads and hepatopancreas. The finding provides an important reference for further understanding of the synthesis and transformation of sex steroid hormones in mollusks.
