摘要 热休克转录因子能够调控真核细胞热休克蛋白的表达,在热休克反应中起重要作用。采用同源克隆和 RACE 方法得到了虾夷扇贝热休克转录因子1基因的 cDNA 全长序列。该序列长1944 bp ,其中5’和3’非编码区分别为54 bp 、318 bp ,编码区序列1572 bp ,包含一个编码523个氨基酸的开放阅读框。经序列比对及生物信息学分析发现,虾夷扇贝热休克转录因子1具有保守的 DNA 结合区域、HR-A /B 和 HR-C 区域,与已知其他物种的热休克转录因子结构相似。该基因在虾夷扇贝6种组织中的实时荧光定量检测结果显示闭壳肌中 mRNA 相对表达量最高,血淋巴中表达量最低。
Abstract: Heat shock transcription factors ,key factors to regulate the transcription of eukaryotic heat shock genes ,play important roles in heat shock response .In this study ,a full length of HSF1 (Heat shock transcription factor 1 ) cDNA was isolated from Yesso scallop Mizuhopecten yessoensis by homologous cloning and RACE method .The full-length was of 1944 bp ,encoding a polypeptide of 523 amino acids .Carried out multiple alignment and bioinformatics analysis ,the predicted amino acid sequence was found to share a conserved DNA-Binding domain ,HR-A /B and HR-C domain ,which showed highly similarities with other HSF1s .The expression of HSF1 mRNA in various tissues was examined using real-time quantitative RT-PCR (QRT-PCR) . HSF1 was expressed in all 6 tissues of the scallop ,and the mRNA expression was the maxinum in adductor muscle and the minimum in hemocytes .
