559 ‘Juvenile Sea Cucumber-Sponge-Macroalage’ Recirculating Aquaculture Technology

This article aims to research a recirculating aquaculture system (RAS) in mariculture .Here we designed and made a RAS with ‘juvenile sea cucumber-sponge-macroalgae’ in a lab to investigate grow th of sea cucumber ,seawater quality ,discharge amount of waste seawater and pollutants in RAS and in control group .During the 30 d experiment ,concentrations of NH3-N ,NO2--N ,NO3--N ,PO3 -4 -P ,and TOC in seawater of sea cucumber tank in RAS were almost similar to those in control group .Moreovel ,fresh body weight of sea cucumber at the end of the experiment reached 75 .5 g (control group) and 84 .6 g (RAS) from body weight of 50 .0 g at the beginning of the experiment .The seawater of 2 .23 m3 (control group) and 0 .6 m3 (RAS) was respectively discharged into coastal ecosystem ;correspondingly ,pollutants also was discharged ,but discharge amount of NH3-N ,NO2--N ,NO3--N ,PO3 -4 -P ,and TOC in RCS was 21% , 25% ,24% ,23% and 59% of those in group control .T he findings indicate that RAS is an environmentally friendly technology system for industrialized mariculture .

567 Effects of Different Fishery Drugs on Activity of Denitrifying Bacteria Pseudomonas stutzeri and Bacillus amyloliquef aciens

Different concentrations of 5 fishery drugs (potassium permanganate ,copper sulfate mixture , chlorine dioxide ,glutaraldehyde and abamectin) were added to the medium of bacteria ,the A600 and the nitrogen concentrations of liquid culture for 24 hours in order to study the influence of fishery drugs on the activity of denitrifying bacteria Pseudomonas stutzeri and Bacillus amyloliquef aciens .The results showed that denitrifying bacteria were most sensitive to potassium permanganate ,glutaraldehyde and chlorine dioxide ,significant decrease in nitrite removal rate ,biomass and the denitrification ability of denitrifying bacteria (P < 0 .05) at 4 mg/L of potassium permanganate ,0 .1 mg/L of glutaraldehyde ,and 0 .3 mg/L of chlorine dioxide (P < 0 .05) ,indicating that these drugs are not mixed in application .However ,the activity of the denitrifying bacteria was not affected by copper sulfate mixture and abamectin which can be mixed in application .B .amyloliquef aciens was most sensitive to potassium permanganate ,copper sulfate mixture ,and chlorine dioxide , significant decrease in removal rate of nitrite at 2 mg/L of potassium permanganate ,0 .25 mg/L of copper sulfate mixture ,and 1 mg/L of chlorine dioxide ( P < 0 .05) , indicating that these drugs are not mixed in the practical application .Glutaraldehyde and abamectin were not found to affect the activity of B .amyloliquef acien ,and be mixed for use .

573 Blood Biochemical Characteristics of Domesticated F 1 in Chinese Sturgeon Acipenser sinensis

Eighteen serum indices including protein ,fat ,glucose ,and inorganic ion levels and enzyme activity were monitored in 3 years old ,8~10 years old ,14~16 years old farmed F1 Chinese sturgeon (Acipenser sinensis) and the Chinese sturgeon with Ⅰ ,Ⅱ and Ⅲ stage gonads domesticated in artificial freshwater environment by automatic biochemical analyzer (type:7170A of Hitachi) to study the biological characteristics of Chinese sturgeon .The results showed that the levels of serum protein including total protein (TP) ,albumin(ALB) ,globulin (GLOB) and Na+ wereshown to be ascendant trend .The levels of serum fat including total cholesterol (CHOL) ,triglyceride (TRIG) ,high density lipoprotein cholesterol (HDLC) and low density lipoprotein cholesterol (LDLC ) were found to be decreased firstly ,and then increased as the time was elapsed while the concentration of serum glucose (GLU) was raised first and then decreased . The maximal activities of serum enzymes including glutamine-oxaloacetate (AST ) , creatine kinase (CK ) , glutamate-pyruvate transaminase (GPT ) , lactate dehydrogenase (LDH ) and alkaline phosphatase (ALP) were observed in the 3 year old individuals of F1 Chinese sturgeon .There were significantly lower 9 blood indices(Cl- ,CK ,ALP ,TP ,ALB ,GLOB ,CHOL ,HDLC and LDLC)in the wild Chinese sturgeon than in 14 ~ 16 years old group of farmed F1 Chinese sturgeon with same developmental gonads (P<0 .05) ,even though the wild Chinese sturgeon showed stage Ⅲ gonads .No significant differences in the other blood indices were found in the wild and farmed Chinese sturgeon (P>0 .05 ) . T he findings indicate that the significant differences in blood biochemistry characteristics are involved in the inherent background variability of these sturgeon individuals ,including exposure history in different habitats ,different developmental gonad and physiological characteristics .

579 Variation in Phytoplankton and Benthic Alga Communities in Sea Cucumber Apostichopus japonicus Culture Ponds

Composition ,density ,biomass ,Shannon-Weiner index ,Pielou’s evenness index ,Margalef’s richness index and McNaugton dominance index of the phytoplankton and benthic alga communities were determined in six 5000 ~ 6000 m2 ponds with depth of 1 .8 ~ 2 .0 m for sea cucumber A postichopus j aponicus culture in coastal Laizihou ,Shandong Province from October ,2011 to September ,2012 .A total of 40 species in 7 phyla of phytoplankon were found ,with the predominant Bacillariophyta ,and 18 benthic alga species in Bacillariophyta .The density was ranged from 7 .810 × 105 to 249 .662 × 105 cell/L ,with biomass of 1 .65~12 .07 mg/L in the phytoplankon and from 4 .124 × 105 to 14 .289 × 105 cell/cm2 ,with biomass of 2 .60~11 .27 mg/cm2 ,in the benthic algae in the ponds .There were Shannon-Weiner index of 2 .09~2 .93 in the phytoplankon and 1 .67~2 .73 in the benthic algae ,and Pielou’s evenness index of 0 .56~0 .78 in the phytoplankon and 0 .48~0 .73 in the benthic algae .The phytoplankon showed Margalef’s richness index from 1 .74 to 2 .42 and the benthic algae from 1 .41 to 1 .83 .The predominant phytoplankton species were found to be changed with time ,but almost no changes were observed in the predominant benthic algae in the year . There were very significant positive correlation between the density of phytoplankton and that of benthic algae in the ponds ,indicating that the species diversity in phytoplankton and benthic algae community is rich and the ecological environment is good in the culture ponds .

585 Comparison of Fatty Acid Composition between Fry and Female Tilapia Oreochromis sp during Spawning Season

The fatty acid composition in muscle of fry and muscle ,hepatopancreas ,abdominal fat and ovary of female tilapia Oreochromis sp were analyzed during spawning season by chemical method and gas chromatograph .The results showed that there were rich in C16:0 ,C16:1 ,C18 :0 ,C18:1n-9 and C18:2n-6 , representing more than 78% in total fatty acid .Fatty acid content in tissues of the tilapia was varied with different growth stages and tissues of the fish .The fry had significantly lower muscular C18:2n-6 content (8 .13% ) than the females (12 .36% ~16 .27% )(P<0 .05)which had 12 .36% ~16 .27% of C18 :2 n-6 in muscle ,abdomen ,and ovary during spawning season(P<0 .05) .The fry had 3 .24% of C20 :4n-6 ,higher than in the other tissues of the females ,next to that in the ovary of the females in which the C20 :4n-6 contents were arranged in various tissues as :ovary > hepatopancreas > muscle> abdominal fat . The contents of C20 :5n-3 and C22 :6n-3 were found to be 0% in the fry ,with maximum (1 .46% and 4 .73% )in the ovary of the females (P< 0 .05) .Ratio of n-3/n-6 < 1 was observed in muscle of fry and muscle , hepatopancreas ,abdominal fat and ovary of the female .The various fatty acid contents were ranged asΣSFA> ΣMUFA> ΣPUFA in muscle of the fry and hepatopancreas and ovary of the females ,and ΣMUFA> ΣSFA> ΣPUFA in muscle and abdominal fat of the females during spawning season .The findings reveal that more C18 :2n-6 content is added into broodstock diet to improve the synthesis of unsaturated fatty acids and to promote gonadal development ,and more C20 :4n-6 content is found in larval diet to improve the juvenile quality .

590 Gene Order,Characteristics and Phylogenetic Analyses of Mitochondrial Genomes in Flatfish Pleuronectiformes

T he gene content of mitochondrial genomes is highly conserved (each encoding 37 genes ) in flatfish Pleuronectiformes .The A + T contents of mitochondrial main coding strands from the suborder Pleuronectoidei ,ranging between 53 .21% (starry flounder Platichthys stellatus ) and 55 .78% (turbot Scophthalmus maximus ) , were lower than those from the suborder Soleoidei . The maximal A + T contents were observed in family Cynoglossidae of suborder Soleoidei ,60 .35% in shortnose tongue-sole Cynoglossus abbreviatus and 60 .59% in half-smooth tongue-sole C .semilaevis .However ,the A + T content from the family Soleidae of suborder Soleoidei (54 .63% in Senegalese sole Solea senegalensis) was found very similar to those of suborder Pleuronectoidei .The ratios of synonymous substitution (K a) to synonymous substitutions (Ks) (K a/Ks) in mitochondrial protein-coding genes were ranged from 0 to 0 .2171 in Hippoglossus ,Verasper and Cynoglossus ,indicating that there is a strong purification (negative) selection in Pleuronectiformes mitochondrial genomes . The comprehensive analysis of 12 Pleuronectiformes mitochondrial gene mutation features show s that nad5 ,nad4 and nad2 genes are ideal molecular markers . Mitochondrial gene order of nine species from the suborder Pleuronectoidei and Senegalese sole from the family Soleidae in suborder Soleoidei is a typical arrangement of vertebrate mitochondrial genomes . However , there are translocation and inversion of tRNA gene (trnQ ) in mitochondrial genomes of shortnose tongue-sole and half-smooth tongue-sole from the family Cynoglossidae of suborder Soleoidei . Mitochondrial genome data support the phylogenetic relationship within Pleuronectiformes at families level : [(Pleuronectidae+ Paralichthyidae)+ Scophthalmidae)]+ Soleidae) + Cynoglossidae . The family Soleidae of suborder Soleoidei was clustered with the suborder Pleuronectoidei firstly (BPN = 100 , BPM = 100) ,followed by clustered with the family Cynoglossidae (BPN = 100 ,BPM = 100) ,indicating that there are two clades in the suborder Soleoidei .Integrated other genomic features ,including A + T content and gene order ,Soleoidei is considered as monophyletic group being questioned ,and need further study .

597 The Feasibility of Green Alga Enteromor pha p roli f ra as Juvenile Sea Cucumber (Apostichopus j aponicus) Feed

The juvenile sea cucumber (Apostichopus japonicus) (body weight 2 .5~3 .5 g) was reared in plastic tanks and fed the diets containing various particle diameters of green alga Enteromorpha prolif ra powder (two commercial feed as control) to confirm the feasibility of the green alga as juvenile sea cucumber feed replacing for brown algae Sargassum pallidum and S .thunbergii at water temperature of (15 .3 ± 2 .8) ℃ for 60 days .Four test feeds were shown to promote growth of the juveniles ,but the best growth was found in the sea cucumber fed the diet containing the green alga powder prepared by 400 mesh w hich acted a function of attractive feeding and led to preventation of the sea cucumber from bad feces . 601 Effect of Stocking Density on Growth of Speckled Spinefoot Siganus guttatus

Speckled spinefoot Siganus guttatus juveniles with body weight of (31 .30 ± 1 .63 ) g were stocked and reared in an indoor tank at the initial stocking density of 2 .48 (group A) ,4 .95 (group B) , 7 .80 (group C) and 10 .20 (group D) kg/m3 at water temperature of 25~27 ℃ for 60 days .The positive relationship between growth and stocking density was found within the rearing biomass of 2 .48~4 .95 kg/m3 .Beyond that density ,there was a negative relationship between growth and stocking density .The grow th parameters revealed that the fish in group B had significantly better grow th than the fish in groups A ,C ,and D ( P <0 .05 ) ,with the maximal body weight of (63 .11 ± 2 .52) g and the maximal body length of (12 .80 ± 0 .98) cm .The fish in group D had the least growth [body weight (50 .73 ± 4 .53) g and body length (11 .90 ± 1 .03) cm] .Food intake were found to be significant decrease with increase in stocking density ,with the maximum in group A (1 .53% ) and the minimum in group D (1 .32% ) .There was significantly lower food conversion ratio in group B than in groups A ,C ,and D (P<0 .05) .It is recommended that the optimal stocking density be of 4 .95 kg/m3 for speckled spinefoot Siganus guttatus .

605 Evaluation of Seawater Eutrophication in Coastal Weihai,Shuangdao Bay Based on Grey Clustering Method

The eutrophication levels were assessed on dissolved oxygen , chemical oxygen demand , inorganic nitrogen ,phosphate and chlorophyll a data collected in coastal Weihai ,Shuangdao Bay ,in June of 2012 ,by a grey cluster method .The results showed that the southern Shuangdao Bay was characterized by eutrophication ,the center Shuangdao Bay by medium-level nutrient ,and the northern Shuangdao Bay by oligotrophication ,which was attributed to the geographic feature of the Bay ,the sea pollution ,and aquaculture activity .It was found that the grey clustering method was more suitable for evaluation of the seawater eutrophication compared with the nutrition condition performance and nutrient index .

609 Immunocytochemical Localization of Neuronal Nitric Oxide Synthase in Brain Tissue of Semi-smooth Sole Cynoglossus semilaevis Günther

The localization of neuronal nitric oxide synthase (nNOS) in the brain tissue of semi-smooth sole Cynoglossus semilaevis Günther was studied using immunohistochemical SP and NADPH-diaphorase staining method .The results showed that the nNOS was found in the cytoplasm of neurons in the cerebra , mostly positive neurons being fusiform , triangle , or round in shape . T he positive nerve fibers in the superficial layer of cerebral cortex were interwoven into a reticulation ,and the positive neurons of cerebel were distributed in the particle layer of cortex ,with many positive nerve fibers and Purkinje cells in the molecular layer .NADPH-d staining showed that there were numerous blue neurons in the cerebra ,and blue neurons ,nerve fibers and Purkinje cells in the cerebel .Western-Blot revealed that there were two protein bands with molecular weight of 118 ku and 80 ku in nNOS in the brain tissue .The findings showed that there was plenty of neuronal nitric oxide synthase in the brain tissue of the sole ,indicating that nNOS can play an important modulatory role in the signal transduction and physiological activity of the fish nervous system .

613 Response of Osmoregulation to Salinity in Mudflat Crab Helice tientsinensis

The hemolymph osmotic regulation was studied in adult mudflat crab ( Helice tientsinensis ) exposed to a salinity of 0 ,10 ,20 ,30 ,40 and 50 for 9 days at 20 °C in aquarium using vapor pressure osmometer .The results showed that there was lower hemolymph osmotic pressure in the mudflat crab exposed to the salinity from 30 to 0 ,higher than that in ambient osmotic pressure .The mudflat crab exposed to the salinity from 30 to 50 had higher hemolymph osmotic pressure ,lower than that in ambient osmotic pressure .The exposure period was found to affect the hemolymph osmotic pressure .There was significant difference in hemolymph osmotic pressure at a salinity of 0 ,10 ,and 50 except 20 ,30 and 40 (P< 0 .05) between 0 h and 3 h after the experiment .The mudflat crab had isosmotic point of 815 .6 mmol/kg ,equivalent to 27 .1 for salinity concentration .The findings indicated that the mudflat crab as an osmoregulator had good regulation of osmotic pressure ability at water temperature of 20 ℃ .
