Abstract:Reactive oxygen species (ROS) play important roles in protection of host from pathogen in in‐vertebrates w hich rely heavily on innate immune mechanisms to defence themselves .Here ,the effects of environmental factors such as water temperature ,salinity and pH on production of ROS by coelomocytes were studied in sea cucumber A postichopus j aponicus maintained under different environmental condi‐tions .Coelomic fluid was extracted at different time points and then ROS production during in vitro phago‐cytosis of yeast cells by coelomocytes was measured 7 d after being acclimatization by the luminol chemilu‐minescence method .The results showed that the ROS production in sea cucumbers held in 6 ℃ and 26 ℃was dramatically increased from the first day and the fifth day compared with the control group (17 ℃) . There were in much more ROS in coelomocytes of the sea cucumbers cultured at salinity of 25 and 35 gen‐erations than in the control group ,while the same went for sea cucumbers kept at salinity of 16 and 40 on the first day .Low pH also led to dramatically enhancement of the production of ROS during phagocytosis . In conclusion ,water temperature ,salinity and pH can influenced the production of ROS by the coelomo‐cytes of sea cucumber during phagocytosis .Which lays a foundation for the further study of the physiolog‐ical and biochemical processes of adaptation of sea cucumber to different environments as well as the innate immune mechanisms in invertebrates .
