Histological Observation of Ovary Development in Xenocypris microlepis

Annual changes in ovaries and oocyte development were histologically observed in Xenocypris microlepis collected monthly in Daoguanhe Reservoir, Hubei Province from Autumn 2007 to August 2008. The development of the ovary underwent six consequent stages while oocytes had five phases, with species specificity from phase Ⅲ. The ovary was under the developing stage before spawning in March. The spawn occurred from April to July, and peaked in May and June. In late July and August, the ovary was in a stage of degeneration after spawning, and then it returned to stageⅡin September and October. From November to February of the following year, the ovary aws in stage Ⅲin the winter. A histological observation of the oocyte development showed that the fish was in a successive process and female seemed to be able to spawn twice, even three times, in one spawning season.

Effects of Ratio among C, N, and P on Growth and Biochemical Compositions in Two Strains of Alga Dunaliella salina

The effects of ratio among C, N, and P on the growth and biochemical compositions in two strains of alga Dunaliella salina called OUN04, and OUN05 were investigated in an orthogonal design test. The maximum cell density of 114.2×10~4 cell/ml, β-carotene content of 100.4 mg/g and chlorophyll-a content of 144.6 mg/g were detected in the alga OUN04 exposed to the media with the ratios of C∶N∶P=12/1/0.05, 12/2/0.1, and 12/1/0.05, respectively, significantly different from those in the control groups. The maximum content of total fatty acids was found to be 86.20% in the OUN04, significantly higher than that in the control group(81.02%). The alga OUN04 had the maximum protein content of 32.44% when the alga was exposed to the medium with the ratios of C∶N∶P=6/1/0.05, insignificantly different from that in the control group. For the alga OUN05, the maximum cell density of 101.2×10~4 cell/ml, β-carotene content of 109.2 mg/g and chlorophyll-a content of 108.7 mg/g were detected in the treated groups with C∶N∶P=6/2/0.05, 6/0.5/0.1, and 12/2/0.1, respectively, significantly different from those in the control groups. The maximum protein content of 34.41% was observed in the C∶N∶P=6/1/0.05 group of, insignificantly different from that in the control group(33.17%).

Improving Enrichment Medium and Culture Conditions in Bacterium Vibrio parahaemolyticus

The bacterium Vibrio parahaemolyticus enrichment medium and culture conditions were optimized by combination of two decisive factors peptone and yeast extract in the experiment designed by statistical software in order to obtain a method for enriching the bacterium. Direct analysis showed that the mixed peptone was found to be the major factor while the yeast extract was considered as the minor in the process of enrichment. The interaction between the two was ignored because of the weak performance. Variance analysis showed that F values of the mixed peptone and yeast extract were more than F_(0.05(1,1)), indicating that the whole values were at the significant level. The optimal medium formula selected by the orthogonal experimental design was described as the following:2% of the mixed peptone(1% tryptone, fish peptone 1%), 0.2% of yeast extract, 3.5% of NaCl,and pH 8.6. And the best results were observed under the culture conditions of (36 ±1) ℃, shaking table at 120 r/min and sealing with 2-layer kraft paper. In the same enrichment time, the concentration of enrichment broth of the bacterium by using the optimized medium and culture conditions was 1.8±0.6 times of the concentration of GB/T 4789.7-2008.

The Effects of Arachidonic Acid(AA)on the Cell Growth and Astaxanthin Content in Alge Haematococcus pluvialis

The effects of arachidonic acid(AA)on the cell growth and astaxanthin content in alge Haematococcus pluvialis were studied in this paper. The results showed that an appropriate concentration of AA(62.5 mg/L)promoted the cell growth of the alga while low concentrations(0.1~12.5 mg/L)and the high concentrations of AA(312.5~1562.5 mg/L)treatments both restrained the cell growth of the alge. AA treatments promoted astaxanthin accumulation, with the increased AA concentrations, the astaxanthin content increased gradually, either. At the rate of 312.5 mg/L AA,the astaxanthin content reached its maximum(3.67 mg/L). There was 48.8% higher in astaxanthin accumulation at 312.5 mg/L treatment compared with the control groups.

Oxygen Consumption and Ammonia Excretion in Jellyfish Rhopilema esculenta with Various Sizes under Different Illumination and Temperature

Oxygen consumption and ammonia excretion were determined by the Winkler method and the Nessler's reagent method in juvenile jellyfish Rhopilema esculenta with various sizes under different ambient illumination, pH 7.9~8.1 and salinity of 25.1. The relationship between dry weight(m)and the bell diameter(D)was found to be represented as the equation:m=0.0003 D~2-0.0007 D+0.0427, r~2=0.9913. There was a negative regression function among oxygen consumption rate(OCR), ammonia excretion rate(AER)and the bell diameter(D):OCR=0.0063 D~2-0.3531 D+6.1533, r~2=0.9751; AER=0.4221 D~2-26.585 D+518.83, r~2= 0.9587. The juvenile jellyfish had significantly greater OCR and AER under light than under darkness(P<0.01). The OCR and AER showed a tendency to increase with the increase in water temperature. The significant increases in AER was only found in 30 ℃ group and other three groups(P<0.01)and the significant increases in OCR were observed in the high water temperature groups except from the 20 ℃ group and the 25 ℃ group(P<0.05). The asphyxia was found at the dissolved oxygen concentration of 0.45 mg/L for the juvenile jellyfish with bell diameter of 15 mm under light than under darkness.

Starvation and Point of No Return in Striped Knifejaw Oplegnathus fasciatus Larvae

Striped knifejaw Oplegnathus fasciatus larvae were starved to determine the point of no return(PNR)at water temperature of 22~24 ℃ and salinity of 29. The survival, and changes in oil globules and yolk-sac volume, growth and swimming behavior in the larvae were observed 0, 3, 6, 9, 12 and 15 days after starvation. The results showed that starvation led to delaying of the use of oil globules, and to the significantly smaller body length than that in the normal larvae in control group(P<0.05). The PNR occurred 5~5.5 days after hatch and the subsistence larvae 0, 3, 6, 9, 12 and 15 d after hatch were death 6.5, 6,5, 4, 6 and 5 days after starvation, respectively.The LC_(50) time of the larvae were 4, 3.5, 3.5, 3.5 d and 3.5 days, respectively, indicating that 9 days was the most sensitive stage to starvation in the early life stage in the larvae. The larvae had shorter body length, bigger head, thinner body and posterior cupped part of brain after being starved for long period. The behavioral responses of the larvae to starvation was divided into three stages:congregating in the surface water, swimming slowly and searching for food; slowly swimming; morphological transformation and resting in the water bottom.

Identification and Drug Sensitive Test of the Pathogen in Acinetobacter Disease from Hybrid Crucian Carp(Carassius auratus gibelio♀× Cyprinus carpio♂)

Five bacteria were isolated from the visceral samples of moribund hybridized crucian carp(Carassius auratus gibelio♀×Cyprinus carpio♂). The ID% and T value of the strain YCS07-04 identified as Acinetobacter baumannii was 99.5% and 0.9 by API 20NE. The artificial infection with three different concentrations showed that the high dosage group with 5.0×10~6 cfu/ml performed high infectivity and caused 100% mortality by injecting in 14 days. Three fish was dead of the bacterium immersing while the healthy fish without clinic disorder was observed in the control group. The isolated bacterium grew well in Nutrient Agar, cloudy with pellicle in nutrient broth, and showed gray white non-hemolytic colonies in rabbit blood agar and colorless transparent colonies on MacConkey agars, but did not live on SS agars. Negative staining under electron microscope revealed that the isolated bacterium was short straight rods with rounded ends in shape and(1.0~1.5)μm×(1.5~2.5)μm in size, becoming spherical in the stationary phase of growth, and to be big irregular filament when gathering. It has rough surface without flagellum, gemma and capsule. The isolated bacterium strain YCS07-04 was sensitive to streptomycin, tobramycin, gentamicin, levofloxacin, lomefloxacin and sulfamethoxazole compounds, and intermediate to tetracycline, while resistant to carbenicillin, ceftazidime,aztreonam and etc.

Rapid Detection of Pathogenic Bacteria of Ascites in Japanese Flounder(Paralichthys olivaceus)by Dot-ELISA

A Dot-Enzyme Linked Immunosorbent Assay(Dot-ELISA)was successfully established for detection of Edwardsiella tarda found in Japanese flounder(Paralichthys olivaceus)by using nitrocellulose membrane as solid carrier. The working concentration of serum samples was 1∶800 dilution and that of the enzyme-labeled goat anti-rabbit IgG was 1∶200 dilution by chessboard assay. The samples with clear and obvious dots were judged as positive reaction. Edwardsiella tarda was positive in the Dot-ELISA, while Aeromonas sobria, A. hydrophila, Vibrio fluvialis, V.alginolyticus , V.anguillarum , Shewanella putrefaciens, Providencia alcalifaciens , Enterobacter sakazakii and Escherichia coli were negative in the assay. The trial indicated that this method was simple, specific, quick, direct, easy extention and application at basic units.

Isolation and Characterization of Haloacid Degrading Bacteria from Marine Sponge Hymeniacidon perleve

Microorganisms capable of producing 2-haloacid dehalogenase have been frequently isolated from the soil, but rarely from the sea. A total of 139 bacterial strains producing the 2-haloacid dehalogenase were enriched and isolated on agar with 2-chloropropionic acid as sole carbon source. Most of the strains showed high salinity tolerance, and were identified as moderately halophilic bacteria. Some of the strains showed the highest salinity tolerance up to 14.6% NaCl. Results of substrate specificity showed that most of the strains degraded a range of organohalogen compounds. These strains, as the first isolation of dehalogenating bacteria from marine sponge, showed good prospects for the further use in biotechnology and environmental field.

Six Kinds of Enzymes in Accessory Gonads of Squid Sepiella maindroni

The activities of six enzymes were measured in accessory gonads of aquid Sepiella maindroni before and after spawn by a kit. The lactic acid dehydrogenase(LDH)(89 U/g prot vs 57 U/g prot in nidamental gland), superoxide dismutase(SOD)(78 U and 47 U/mg prot vs 51 U and 32 U/mg prot), acid phosphatase(ACP), peroxidase(POD)and catalase(CAT)were found to decrease in activities after spawning.

The Acute Toxicity of Non-ionized Ammonia to Juvenile Goldfish Carassius auratus

The acute toxicity of non-ionized ammonia to juvenile goldfish Carassius auratus(5.0±0.61) cm in body length, and(12.17±1.57) g in body weight was studied via a general method. It was found that the 24 h CL_(50), and 48 h LC_(50) values(95% confidence interval)of nitrite-N to the juveniles were 0.454(0.393~0.515), and 0.267(0.213~0.322) mg/L, with security concentration of 0.028 mg/L, indicating that the juveniles has lower toleration ability.
