405 Antibacterial Activity of Chinese Herbal Medicine against Pathogenic Aeromonas caviae in Cultured Giant Salamander Andrias davidianus in vitro

Chinese herbal medicines against pathogenic Aeromonas caviae were screened for cultured giant salamander Andrias davidianus in vitro .Effects of pH values on inhibition against A .caviae by Chinese herbal medicines were studied by agar plate diffusion method ,including the minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) of single drug and the compounds against pathogenic A .caviae by agar double dilution method .It was found that 200 kinds of Chinese herbal medicines had different antibacterial effects on the pathogenic bacterium ,135 kinds showing to some extent antibacterial activity ,and 65 kinds showing no antibacterial activity .Terminalia chebula Retz ,Rhus chinensis Mil ,Punica granate ,Rheum palmatum and Sophora j aponica had the minimum MIC and the minimum MBC of less than 1 .0 mg/mL . Terminalia chebula Retz ,Rhus chinensis Mil ,Schisandra chinensis and Dendranthema morifolium showed inhibitory and bactericidal actions against A .caviae at pH near acidity .The findings provide a theoretical basis with exploitation of efficient ,safe and environmental friend drugs against pathogenic bacteria in giant salamander .

413 An Experiment on Polyculture of Pacific White Leg Shrimp Litopenaeus vannamei with Malabar Grouper Epinephelus malabaricus

Pacific white leg shrimp Litopenaeus vannamei with body length of 2-3 cm ,4-5 cm and 8-9 cm was reared in concrete tanks of each 2 .5 m × 4 .0 m × 2 .0 m at a stocking rate of 3000 thousand individuals/hm2 for 10 days when malabar grouper with body length of 12 .3-13 .6 cm and 17 .6-18 .8 cm was stocked into the tanks at a rate of 5 fish per tank and reared for 60 days with duplication .In the control tanks ,only Pacific white leg shrimp was stocked and cultured .The results showed that there were significantly lower body weight gain and significantly higher survival for 2-3 cm and 4-5 cm shrimps in monoculture than those in the polyculture(P<0 .05) ,the maximal body weight gain in polyculture 2 -3 cm shrimps with body length of 17 .6-18 .8 cm(7 .88 g)and 4-5 cm shrimps with body length of 17 .6-18 .8 cm (7 .67 g)groups .No significant differences in survival were found in 8 -9 cm shrimp in both monoculture and polyculture (P>0 .05) .The body weight gain and survival of shrimp were affected by the size of Pacific white leg shrimp ,culture model and their interactions (P<0 .01) .In polyculture ,there was no significant effect of the body length of shrimp on body weight gain of malabar grouper (P>0 .05) ,the body weight gain being significant higher in body length of 17 .6-18 .8 cm groups than in body length of 12 .3-13 .6 cm groups (P<0 .05) .The malabar grouper showed less predation of the larger shrimp with higher survival in 8-9 cm shrimps with body length of 17 .6-18 .8 cm malabar grouper group ,indicating that there was very significant effect of sizes of shrimp and malabar grouper on growth and predation of malabar grouper (P< 0 .01) .The benefit analysis showed that the profit of Pacific white leg shrimp polyculture was significantly higher than that in monoculture (P< 0 .05) ,the profit being significantly higher in 4-5 cm shrimps polycultured with body length of 17 .6 -18 .8 cm fish ,4 -5 cm shrimps with body length of 12 .3-13 .6 cm fish ,and 2-3 cm shrimps with body length of 17 .6-18 .8 cm fish models than in the other models .The 8 -9 cm shrimp showed significantly less profit than others ,and malabar grouper was recommended to be stocked into the tanks when Pacific white leg shrimp had body length of4-5cm.

418 Antioxidant Defenses and Lysozyme Activity in Chinese Mitten-handed Crab Eriochei r sinensis Farmed in Organic Farming Pattern

The activities of lysozyme ,superoxide dismutase (SOD) ,catalase (CAT ) ,and glutathione peroxidase (GSH‐Px) ,and contents of malondialdehyde (MDA) were studied in gill ,hepatopancreas and muscle of Chinese mitten‐handed carb (Eriocheir sinensis) with body weight of 165-175 g farmed under organic and traditional farming patterns . The results showed that the activities of SOD and CAT in hepatopancreas and muscle were significantly higher in the crab farmed in organic pattern than in traditional pattern (P <0.05) ,without significant difference in the activity of SOD and CAT in gills (P>0 .05) .The activity of GSH‐Px was found to be much higher in gill ,hepatopancreas and muscle of Chinese mitten‐handed crab cultured in organic pattern than in traditional pattern (P <0 .05) .There was significantly lower content of MDA in gill , hepatopancreas and muscle of Chinese mitten‐handed crab cultured in organic pattern than in traditional pattern (P <0 .05) .Lysozyme activitiy was much higher in hepatopancreas of Chinese mitten‐handed crab cultured in organic pattern than in traditional pattern (P <0 .05) .The findings indicate that Chinese mitten‐handed crab cultured in the organic pattern show stronger ability of antioxidant defenses and resistance to diseases than the crab farmed in the traditional pattern .

423 Optimal Dietary Enteromorpha prolifera in turbot (Scophthalmus maximus)

To study the application of dietary filamentous alga Enteromorpha prolifera in juvenile turbot , the experiment was conducted to determine the effects of dietary E .prolifera on growth ,feeding and nonspecific immune responses .The turbot juveniles with initial weight (6 .05 ± 0 .03)g were fed five different diets containing green alga E .p roli f era at 0% (E1 ,control group) ,5% (E2) ,10% (E3) ,15%(E4) and 20% (E5) for 42 days .Fish were randomly sampled for analysis of the weight gain rate ,specific grow th rate , feed efficiency rate , food ingestion , macrophage phagocytic activity and macrophage respiratory burst activity .The results showed that the specific growth rate ,growth rate and feed efficiency rate were significantly lower in the E5 group than those in the test group (E1) (P<0 .05) .There was no significant difference in food ingestion among every group (P> 0 .05) .The macrophage proliferation activity and macrophage respiratory burst activity were significantly higher in E 2 group than in the other groups (P<0 .05) .Under the experimental conditions ,there were significantly increase in growth and non‐specific immunity in the turbot fed the diet containing 5% E .prolifera .However ,20% E .prolifera dietary had the negative effects on the grow th and non‐specific immunity in turbot .

428 Identification and Antibiotic susceptibility of Pathogenic Bacteriium Aeromonas hydrophilia from Diseased Scortum barcoo

Two pathogenic bacteria (SS1495 ,and SS1496) were isolated from diseased Scortum barcoo in an aquafarm which was located in Sanshui district of Foshan city , Guangdong province . They were identified by microbial biochemical identification tubes and 16S rDNA sequence analysis ,and the artificial infection experiment and antibiotic susceptibility analysis were performed .The results showed that these two bacteria were facultative anaerobes which were gram negative ,rod in shape ,and motile .They had positive indole ,methyl red and V‐P characteristics ,and utilized glucose ,maltose and mannitol .According to“Bergey’s Manual of Determinative Bacteriology”,the isolates were identified to be Aeromonas hydrophilia based on the morphology ,physiological and biochemical characteristics of the bacteria .The 16S rDNA sequences of the isolates showed a high similarity to that of A .hydrophilia via BLAST network services in NCBI .The median lethal doses for the isolates to healthy S .barcoo were 7 .8 × 104 cfu/g and 3 .9 × 104 cfu/g ,respectively ,indicating they had strong pathogenicity to S .barcoo .The isolates were highly susceptible to cefotaxime ,ciprofloxacin , norfloxacin ,and levofloxacin ,intermediately susceptible to phytomycin ,cephazoline and neomycin ,and resistant to tetracycline ,rifampicin and lincomycin .

433 Histopathological Changes in Juvenile Grass Carp with Parasite Ichthyophthirius

Histopathological changes were observed in brain ,heart ,gill ,fin ,head kidney ,midkidney , premaxilla ,skin and viscus in juvenile grass carp Ctenopharyngodon idellus infected with protozoon Ichthyophthirius sp .The protozoon was found in various organs included gill filaments ,premaxilla , pectoral fins and dorsal fins ,and there was congestion in the brain ,and the auricle ,ventricule and arterial balls of the heart .There was congestion in the gill filaments with inflammatory cells infiltration ,the disappeared partly gill lamella structure ,and respiratory epithelial cells .The congestion in the head kidney , the fatty degeneration and hyalinization of epithelial cells of renal tubuls and the connecting tubule ,and enlargement of glomerula led to the disappearance of renal follicle ,with congestion in renal interstitium . The collagen fiber of dense layer in abdominal subcutaneous tissue showed part hyaline degeneration ,the congestion vein ,the intestinal mucous epithelial cell shed partly ,and the almost dissolved epithelial cells . The serious congestion was observed in the hepatopancreas ,and venous congestion appeared in the liver sinusoids and spleen .

438 Optimization of Ultrasound-assisted Autolysis and Exogenous Hydrolysis in Jinjiang Oyster(Crassostrea rivularis)

Ultrasound and protease can improve yields of short peptide by destroying the structure of protein .The effect of ultrasound power and time on autolysis of Jinjiang oyster (Crassostrea rivularis) was invesigated firstly to obtain high short peptide content considering hydrolysis degree and total volatile basic nitrogen .Then single‐factor experiment was conducted to evaluate effect of alkaline hydrolysis on the ultrasound‐assisted autolysis hydrolysate .The optimum ultrasound pretreated conditions were ascertained as 400 W ,and 10 min ,with short peptide yield of up to (88 .75 ± 1 .16) mg/mL ,2 .12 times as the control group .The total volatile basic nitrogen level was reduced to (79 ± 2) μg/g with hydrolysis degree of (30.38 ± 0 .05 )% . After ultrasound‐assisted autolysis , the hydrolysis conditions with alkaline were optimized ,and the results showed that enzyme concentration of 5 kU/g ,temperature 55 ℃ ,and for 4 h led to short peptide content of above 100 mg/mL ,with hydrolysis degree of (35 .95 ± 0 .05)% and total volatile basic nitrogen of (178 ± 6) μg/g ,which was in an acceptable range .The findings indicate that using ultrasound‐assisted autolysis with exogenous hydrolysis can significantly improve efficiency of enzymolysis .

444 Acute Toxicity of Propylene Oxide,Acrylic Acid and Isoprene to Juvenile Sea Cucumber Apostichopus j aponicus

Acute toxicity of propylene oxide , acrylic acid and isoprene to juvenile sea cucumber A postichopus j aponicus with body weight of (1 .30 ± 0 .62) g was studied at water temperature of 16—19 ℃ by static bioassay .It was found that LC50 of propylene oxide ,acrylic acid and isoprene to juvenile sea cucumber was 181 .53 ,117 .63 and 70 .40 mg/L for 24 h ,148 .91 ,109 .81 and 68 .28 mg/L for 48 h , 133.62 ,102 .70 and 67 .25 mg/L for 72 h ,and 98 .89 ,100 .34 and 66 .55 mg/L for 96 h ,with safe concentration of 9 .89 ,10 .03 and 6 .66 mg/L ,respectively ,indicating the descendant toxicity order as isoprene(moderate toxicity )> propylene oxide (moderate toxicity )> acrylic acid (low toxicity ) . The findings may provide the reference to ecological toxicology research of propylene oxide ,acrylic acid and isoprene to marine organisms .

448 Changes in Activity and Distribution of Cathepsin L-Like Proteases in Autolytic Octopus (Octopus ocellatus)

The contents of water‐soluble total proteins and collagen proteins ,as well as the activities of endogenous total proteases and cathepsin L‐like proteases were determined in arm muscle of the autolytic octopus induced by ultraviolet (UV ) . Results showed that the octopus became insensitive after UV irradiation .The body of octopus became softened ,and mucoid degeneration took place in epidermis and subcutaneous tissue .The changes in external appearance indicated that autolysis took place .Along with autolysis ,the contents of water‐soluble total proteins and collagen proteins were found to be increased , indicating degradation of structural proteins .At the same time ,the activities of endogenous total proteases and cathepsin L‐like proteases tissue were increased in tissues in wnit mass .The distribution of cathepsin L‐like proteases in arm muscle of the autolytic octopus was in homogeneous . The stained spots of cathepsin L‐like proteases were dense in myocardial trabeculation ,but were relatively sparse in muscle fibers .Along with the progress of autolysis ,the stained spots of cathepsin L‐like proteases were scattered and eventually disappeared ,indicating the dispersion of cathepsin L‐like proteases in tissue .The findings indicated that the endogenous proteases , cathepsin L‐like proteases as representive , took part in the autolysis of octopus .

453 Effects of Partial Replacement of Fish Oil by Soybean Oil on Serum Lipid Metabolism and Related Enzyme Activities in Chinese Striped-necked Turtle Mauremys sinensis

In order to study the effects of partial replacement of fish oil by soybean oil on serum lipid metabolism and the activity of correlated lipases . Four‐year old female Chinese striped‐necked turtle Mauremys sinensis [body weight(1557 ± 307) g] were randomly divided into four groups according to and fed the diets containing various proportions of fish oil (F) and soybean oil (S) including 0S∶3F (S∶F , 0∶3) ,1S∶2F (S∶F ,1∶2) ,2S∶1F(S∶ F ,2∶1) and 3S∶0F (S∶ F ,3∶0) for eight months .Six turtles were randomly selected from each group and determined for the content of triglycerides (TG) ,total cholesterol (TC) ,high density lipoprotein cholesterol (HDL‐C) and low density lipoprotein cholesterol (LDL‐C) in blood collected from the heart and the activity of lipoprotein lipase (LPL ) ,hepatic lipase (HL) and hormone sensitive lipase (HSL) in liver ,and pancreatic lipase (PL) in intestine .The results showed that the content of TC was significantly higher in 1S∶2F and 3S∶0F than that in 0S∶3F (P<0.05);the content of LDL‐C was significantly higher in 3S ∶ 0F than in 0S ∶ 3F ( P < 0 .05 ) , no significant differences in the other groups . The maximal activity of the total lipase (LPL + HL ) was observed in 2S∶1F ,significantly higher than that in the 0S∶3F group(P < 0 .05) and almost the same in HSL in 2S ∶ 1F to 0S ∶ 3F compared to the rest groups of HSL . There was no was not significant differentce in PL in the group of 2S∶1F from that in 0S∶3F ,but both being lower than that in the other groups .In totally of enzyme activity ,the group of 2S∶1F was also the suitable group .Above all ,the substitution of fish oil by soybean oil in the turtle could be worked ,with the suitable ratio of 2S∶1F .
