63 A Comparative Study on Surface Morphology of Sperm and Mature Egg in Three Species Flatfish

The surface morphological structures of sperm and mature egg of three species flatfish Japanese flounder Paralichthys olivaceus ,turbot Scophthalmus maximus L,and Verasper moseri were studied by scanning electron microscopy. Comparing results demonstrated that there are four obvious differences in sperm surface structures among three species flatfish,including the location of nucleus anterior pit,the shape of sperm head,the number of mitochondria in midpiece and length of the flagellum. But in the surface morphology of mature egg of three species flatfish,it is similar essentially among Japanese flounder and turbot,excepting stripe stars flounder. Four structure characteristics of the shell membrane are observed in V. moseri ,they are a rough outer surface,various microspores in size,diameter as well as the most thick shell membrane(3.87±0.24) μm which is twice thicker than Japanses flounder and turbot and spiral pipe layer in micropyle only for 4-6 layers about half of Japanses Flounder and turbot. It was concluded that the difference was obvious in the surface morphology of sperm and mature egg between three species of Flatfish,and stripe stars flounder as genetic distance of the farthest Pleuronectidae fish,have significant level.

69 Effects of Nutritional Factors on Growth in Alga Heterosigma akashiwo

The effects of N,P,microelements and vitamins on the growth of alga Heterosigma akashiwo were investigated by orthogonal designed test. The results showed that the effect range of various nutritional factors was in a descent order ranged as the following:P>vitamins>N>microelements in exponential and stationary phase and N>P>vitamins>microelements in decline phase. In exponential phase,the optimal concentration of N,P,microelements and vitamins was found to be 880,18,10.55 and 0.32 μmol/L,and 2640,108,21.1,and 0.96 μmol/L in stable and decline phases,respectively. The nutritional factors had significant effect on growth of the alga ( P <0.01) during various phases and the effect of the nutritional factors on the alga bloom were discussed in detail.

74 Habit and Adaptability of Common Octopus Octopus variabilis to Certain Environmental Factors

The habit and adaptability of common octopus Octopus variabilis to water temperature,salinity and pH were studied in a tank with a shelter of tiles,bricks,rocks and plastic tubes. During the experiment,the water temperature was regulated by an electric heater and ice packs,the pH values were controlled by addition of NaOH and HCl and the salinity was maintained by various proportions of fresh water and NaCl. The results showed that the common octopus tended to inhabit under the shelters of tiles,bricks and rocks and rested in the mud seafloor at a water temperature of 10~31 ℃,optimally ranging from 12 to 27 ℃,a salinity of 16.3~27.3,alternatively from 18.3 to 24.3,and the pH from 6.2 to 9.7 with the optimal range of 6.7~9.2.

79 The Optimal Culture Conditions of Bacterium Vibrio harveyi WG1702 from Clam Meretrix meretrix

A bacterial strain WG1702( Vibrio harveyi) was isolated from diseased clam Meretrix meretrix during epidemic breakdown in Lvsi harbor,Jiangsu province,in September of 2007. The optimization of culture conditions showed that the strain WG1702 reached the peak growth in 32 h culture at temperature of 28 ℃,initial pH of 7,salinity of 40,and aeration ratio of 60 ml/150 ml. The metal ions such as NH_4~+,Fe~(2+) ,and Fe~(3+) were used as a cell growth enhancer,glucose as a carbon source and peptone as a nitrogen source. The orthogonal experiment for different age bacteria proved that the best growth was observed under the combination of the pH 7.11,temperature 28 ℃,and 24 h age bacteria. The variance analysis revealed that pH value had significantly influence on the growth of the bacterial strain.

83 Effects of Shell Size Characters on Live Weight in Sea Urchin Strongylocentrotus intermedius

The correlation coefficients were measured and calculated among shell volume( x_1) ,test diameter( x_2) ,shell height( x_3) ,and the live body weight( y ) in 66 samples of sea urchin Strongylocentrotus intermedius during 13 months. The results showed that 3 correlation coefficients between each shell character and the live body weight were all very significant differences( P <0.01) . The test diameter( x_2) showed predominant effect( P_2=0.778) on the live body weight. The shell volume( x_1) gave slightly direct effect and indirect effect on the live body weight. Depended on the check on path analysis,shell volume( x_1) had no significant in multiple regression equation. It was clear from the results of high correlation index(R~2=0.8978) that the truthful relationship between the independent variables( x_2 and x_3) and the dependent variable( Y ) was revealed by the path coefficient analysis. The multiple regression equation to estimate live body weight as y -21.78+1.035 x_1+0.188 x_2. This paper provides theoretical evidence and perfect measure target for breeding of sea urchins.

87 Histology and Ultrastructure of the Nidamental Gland of Cuttlefish( Sepiella maindroni )

The histology and ultrastructure of the nidamental gland of cuttlefish Sepiella maindroni were studied using light microscopy and electron microscopy. The nidmental gland was found to consist of two parts:an nidamental wall and many lamellae containing two types of cell(cells TypeⅠand cells typeⅡ) . The main function of Cells TypeⅠ was to secret most of the granules made of glucoprotein,and cells TypeⅡ did not secrete granule and maybe have a function of supporting the cells type.

91 Extraction of Polysaccharides from Razor Clam Sinonovacula constricta and Their Antioxidant Activity

The crude product of polysaccharides was isolated from fresh razor clam Sinonovacula constricta chopper added water by ultrasonic extraction and further purified by DEAE ion-exchange chromatography and Sephadex G-75 chromatography. The antioxidant activity of the polysaccharide substances was determined by fenton system and Pyrogallol oxidation. The results showed that the polysaccharide rate of ultrasonic extraction was 19.93%. The razor clam polysaccharides had effect on scavenging hydroxyl free radicals,and the scavenging hydroxyl free radical ability was increased up to 94.76% and scavenging superoxide anion radical ability was up to 83.08%.

95 Changes in Fatty Acid Composition in Muscle of Largefin Longbarbel Catfish Mystu macropterus

Fatty acid composition was analyzed in the muscle of largefin longbarbel catfish Mystus macropterus by gas capillary chromatography. The results showed that there were 22 fatty acids in the muscles,the predominant fatty acids,in terms of relative proportion,being C18:1n-9(22.38%~37.57%) ,followed by C16:0(18.86%~23.95%) ,C16:1n-7(7.00%~11.52%) ,and C18:2n-6(7.46%~9.97%) . The proportion of EPA and DHA were relatively high,accounting for 3.74% and 5.22%,respectively. Eight saturated fatty acids (SFA) ,6 monounsaturated fatty acid(MUFA) and 8 polyunsaturated fatty acids (PUFA) were found in the muscles. Fatty acid composition showed difference in the muscle in the pre-spawning and post-spawning.

99 Effects of Supplementation of Methionine in Soybean Meal-based Diets on Growth of Yellow Catfish Pelteobagrus vachelli

An 8 week feeding experiment was conducted to evaluate the influence of supplementation of methionine(0,0.2%,and 0.4%) in soybean meal-based diets on growth of yellow catfish Pelteobagrus vachelli . There were significantly higher weight gain in the fish fed the diets containing 0.2%,and 0.4% of methionine than in the fish fed the diet containing 0% of methionine( P <0.05) . The peak weight gain(176.22±3.56) % and significantly higher survival rate were observed in the fish fed the diets containing 0.2% of methionine( P <0.05) . There were no significant differences in feed conversion ratio and food consumption( P >0.05) among the experimental groups. Between every groups,even though the fish fed the diet supplemented with 0.2% of methionine had 12.50% higher food consumption and lowest diet ratio in the contorl group (2.60±0.13) . Therefore,under this condition,it is suggested that the optimal supplementation of methionine in the yellow catfish diets be 0.2%~0.4%.

102 Inhibitory Effect of Three Disinfectors on Two Pathogenic Bacteria in Aquatic Animals in a Carrier Quantitative Germicidal Test

The bactericidal performance of Tetrakis Hydroxymethyl Phosphonium Sulfate (THPS),2-Bromo-2-nitro-1.3-Propanediol (BNP) and formalin was studied in a carrier quantitative germicidal test. The results showed that the average inhibition effect of THPS on Aeromonas hydrophila strain BSK-10 was 100% at the concentration of 0.500 μl/L. The average inhibition effect of THPS on the strain GYC1108-1 was 100% at the concentration of 500 μl/L. The average inhibition effect of THPS on Edwarelsiella (Et) was found to be 100% at the concentration of 1.00 μl/L. The average rate of inhibition effect of BNP on BSK-10 was 100% at the concentration of 16 mg/L. The average inhibition effect of BNP on GYC1108-1 was 100% at the concentration of 8 mg/L. The average rate of inhibition effect of BNP on the strain Et was 100% at the concentration of 8 mg/L. The results showed that the germicidal efficacy of THPS was stronger than that of formalin and BNP in the same concentration.

106 Artificial Breeding of Yellow Catfish Peltebagrus fulvidraco Richardson in a Large-scale
A new incubation procedure with high production of fry was established according to the biological properties of spawning and larvae of yellow catfish Peltebagrus fulvidraco by the experiments conducted in Zhongxin Freshwater Fisheries Co.,Ltd in Dengta City from 2005 to 2007. Comparing with production of 2005,significant increase was shown in 2006 and 2007 by the hatching procedure. Total of 36 465 broodstocks and 27 010 broodstocks were induced to spawn by artificial propagation,and 109.68 million larvae and 106.834 million larvae were obtained in 2006 and 2007,respectively,average 7821 individuals per broodstock,the highest on the record. Furthermore,the healthy matured broodstocks and good water quality are important factors in the artificial breeeding.

109 Acute Toxicity of Three Heavy Metal Ions to Rose Bitterling Rhodeus ocellatus Fry

Acute toxicity of Hg~(2+) ,Pb~(2+) and Zn~(2+) to rose bitterling Rhodeus ocellatus fry was studied by static bioassay test. The LC_(50) of Hg~(2+) for 24 h,48 h,72 h,and 96 h was found to be 0.6123,0.4734, 0.3921,and 0.3621 mg/L for the fry,respectively;LC_(50) of Pb~(2+) 14.1421,13.7405,13.5261,and 13.1867 mg/L and LC_(50) of Zn~(2+) 24.4688,20.3542,19.5449,and 17.8608 mg/L,respectively. The safety concentrations of Hg~(2+) ,Pb~(2+) and Zn~(2+) were found to be 0.0362,1.3186,and 1.7861 mg/L, respectively.

112 Bacteriostasis of Common Microalga Used in a Shrimp Hatchery

Inhibition of pathogenic bacteria Vibro anguillarum and Pseudomonas sp.by algae Isochrysis galbana,Phaeodactylum tricornutum and Tetraselmis chuii was studied in this experiment. I. galbana and P. tricornutum 's aqueous phase,and I. galbana,P. tricornutum and T. chuii 's organic phase were found to inhibit the pathogenic strains of V. anguillarum. It was found that I. galbana,P. tricornutum and T. chuii' s organic phase showed inhibition of Pseudomonas sp . I. galbana,P. tricornutum and T. chuii had the maximal inhibitory activity on the pathogens when the algae were kept at density of about 10,000 cells/ml in larval shrimp rearing water.
