Effects of Different Hormones on Artificial Propagation in Cultured Russian Sturgeon,Acipenser gueldenstaedti

Three kinds of hormones were evaluated based on the results of artificial propagation in cultured Russian sturgeon .Injection of carp pituitary ,L HRH-A2 and L HRH-A2 +DOM induced cultured sturgeon brood fish mature and spawn .Among them ,the combination of LHRH-A2 +DOM was the most effective hormones based on results including latency time (22-38 h) ,success rate(80% ) ,fertilization rate (83 .2 ± 3 .46)% and hatching rate(83 .2 ± 3 .46)% .There was no significant difference in induction of sperm and quality among difference experimental groups .Duration of sperm quick movement was 58-115 s ,the life span of sperm was 130-235 s ,and the sperm density was 0 .1 × 109 -2 .21 × 109/mL .

Effects of Anti-stress Agents on Serum Biochemical Parameters and Hepatic Antioxidant Parameters in Wuchang Bream (Megalobrama amblycephala) during Long-distance Transportation

The serum cortisol (COR) ,levels of 3 ,5 ,3′ - triiodothyronine (T3) ,complement 3 (C3) , complement 4 (C4) ,hepatic malondialdehyde (MDA) ,activity of superoxide dismutase (SOD) ,and total antioxidant capacity (T-AOC) were determined in Wuchang bream (Megalobrama amblycephala) with initial body weight of (600 ± 12) g held in 1m3 tanks of 750 fish fed the diets containing 20 mg/L vitamin C , glutamine or anti-stress compund (anti-stress agents ) (without addition as control group ) during long-distance transportation period of 0 h ,4 h ,8 h ,and 12 h to evaluate the effect of anti-stress agents on the serum biochemical parameters and hepatic antioxidant abilities during the long-distance transportation period .The results showed that the anti-stress agents led to significantly (P<0 .05) decrease in serum cortisol levels and hepatopancreas MDA levels ,and to significantly (P <0 .05) increase in serum C3 ,C4 , hepatic SOD ,and T-AOC compared with the control group ,indicating that the anti-stress agents was used to regulate biochemical parameters and antioxidant ability .Based on the serum levels of COR ,T3 ,C3 , C4 ,and hepatic MDA ,SOD activity ,and T-AOC ,the antioxidant ability order of the anti-stress agents was ranged as vitamin C>glutamine>anti-stress compound .

Effects of Beneze,Toluene,Ethylbenzene,and Xylene (BTEX) on Lipid Peroxidation in Sea Cucumber Apostichopus j aponicus

T he effects of various concentrations and exposure time of beneze ,toluene ,ethylbenzene ,and xylene (BTEX) on lipid peroxidation and malonaldehyde (MDA) content were studied in the coelomic fluid of sea cucumber A postichopus j aponicus with body weight of (5 .00 ± 0 .25) g exposed to the BTEX for 0 h ,24 h ,36 h ,48 h ,60 h ,72 h ,84 h and 96 h at water temperature of (17 ± 0 .5) ℃ and a salinity of 32 ± 0 .5 .Results showed that there were no significant changes in MDA content between low concentration benzene (0 .81 mg/L ) and toluene (0 .47 mg/L ) and the control group in the coelomic fluid of the sea cucumber during the experiment .In mid and high concentration of benzene (4 .06 mg/L and 20 .32 mg/L) and toluene (2 .33 mg/L and 11 .66 mg/L) ,and three concentrations of p-xylene (0 .28 mg/L ,1 .38 mg/L and 6 .92 mg/L) ,m-xylene (0 .22 mg/L ,1 .09 mg/L and 5 .44 mg/L) ,o-xylene (0 .16 mg/L ,0 .81 mg/L and 4 .05 mg/L) and ethylbenzen (0 .35 mg/L ,1 .74 mg/L and 8 .68 mg/L) ,however ,the MDA contents were found to be gradually decreased with increase in exposure time ,being inhibited significantly by both p-xylene and m-xylene contents .The findings indicate that BTEX might cause lipid peroxidation injury of the sea cucumber and that the MDA content might be used as biomarkers to evaluate the effects of BTEX on marine environment .

A Rapid Sensitive Detection Method of Pathogen Edwardsiella tarda

Edw ardsiella tarda as an etiological agent of edwardsiellosis ,a serious systematic disease ,is responsible for large economic losses in aquaculture .Recently ,a number of E .tarda strains isolated have exhibited resistance to multiple antibiotics ,and lead to difficulty to effective clinic therapy .In order to exert effective and in-time therapeutic measures against edwardsiellosis ,rapid antimicrobial susceptibility testing methods are urgently needed to be developed for scientific and reasonable selection of antibiotics in therapy .Based on MH broth ,enrichment broth for E.tarda strain JF-1 was optimized by addition of fish peptone and inorganic salt ions ,and Alamar Blue was employed as the color indicator ,and 11 kinds of antibiotics were inoculated into the 96-well microplates at 8 different concentrations by doubling dilution for construction of rapid antibiotic sensitivity testing (AST) plate which contained AST region ,growth control region and blank region .Compared with the color-developing method and spectrophotometric assay ,the strain JF-1 suspension at 1 × 105 cells/mL as optimal inoculation concentration was inoculated into the prepared rapid AST plate ,and detected by a spectrophotometric assay and tube dilution method for AST in parallel as comparison .12 h post inoculation , the MIC data obtained by color-developing method on the rapid AST plate was in accordance with the results acquired by spectrophotometric assay , and showed a little deviation with the results obtained by tube dilution method at 24 h post inoculation . The results showed that the rapid AST method developed based on color-developing was well supported by spectrophotometric assay and tube dilution method ,although there is a little deviation between the results obtained from tube dilution method and the rapid AST method . The wells in the microplate have intrinsically a large surface-to-volume (S/V) ratio resulting in a higher oxygen dissolving efficiency ,which would greatly benefit the bacterial propagation and show a less susceptibility to a certain antibiotic concentration compared to the tube dilution method .The findings could be further applied to the establishment of rapid AST kit for E .tarda ,and provides a basic reference for development of simple , accurate ,effective and rapid AST approaches for aquaculture .

Identification of Heterozygous Clone and Genetic Analysis of Growth Traits in Japanese Flounder

In this study ,mitotic gynogenetic diploid Japanese flounder (Paralichthys olivaceus) were produced by activation of ultraviolet irradiated sperm of red sea bream ( Pagrosomus major ) , and by hydrostatic pressure treatment to block the first mitotic division to identity the heterozygous clone .Nine heterozygous clones (HC1-HC9) of Japanese flounder were produced by crossing nine males with nine females after these diploid flounder were cultivated until sexual maturity .Simultaneously ,two homozygous clones (C1-C2) were prepared by inducing two females to carry out meiotic gynogenesis .A set of 22 microsatellite markers with high polymorphism were selected from the second genetic linkage maps of Japanese flounder .Genetic status of these heterozygous clones was identified and the association between the number of heterozygous loci at alleles and phenotypic value for grow th traits of different heterozygous clones were analyzed .The results showed that the genotypes of heterozygous clone offsprings were identical and the combination of parental alleles ,indicating the successful development of heterozygous clones .The association analysis revealed that the minimal phenotypic values for growth traits were found in 2 homozygous clones including zero heterozygous loci .In 9 heterozygous clones ,the phenotypic value for growth traits of corresponding heterozygous clone was first increased and then decreased with increase in the number of heterozygous loci .The maximal phenotypic value for growth traits were found in HC5 containing 5 heterozygous loci .The HC9 had the maximum number of heterozygous loci ,but its phenotypic value for growth traits was not the optimum .Thus ,there was to some extent relationship between the number of heterozygous loci at alleles and phenotypic value for growth traits , which would provide theoretical references for parent selection of heterozygous clone and genetic improvement of objective traits in Japanese flounder in the future .

Effects of Hg on Three Defense Related Enzyme Activities in Early Developmental Stages of Pacific Oyster Crassostrea giga

The effects of heavy metal Hg on three defense related enzyme activities were investigated in embryos and larvae of Pacific oyster Crassostrea giga to elucidate the effects of heavy metals on physiological and biochemical indexes and toxic mechanism during early developmental stages of bivalves . The results showed that the acid phosphatase (ACP) and alkaline phosphatase (ALP) activities were found to be reduced significantly (P<0 .01) in embryos and larvae exposed to Hg concentrations of 3 μg/L ,30μg/L and 300 μg/L for 2 h (morula stage) ,5 h (gastrula stage) ,8 h (trochophore larval stage) ,and 20 h (D-shaped veliger larval stage) compared with those in control group .The peroxidase (POD) activities of embryos and larvae were also found to be inhibited significantly (P<0 .05) in the groups of 3μg/L and 30μg/L compared with the control group ,while the POD activity were increased in 30 μg/L group (P<0 .01) .In control group ,three enzyme activities were changed with the development of Pacific oyster , with increases in activity in morula stag ,then decrease ,the minimum in gastrula and trochophore stages and increase in veliger larvae .The findings indicate that the toxic effects of metal on embryos and larvae of bivalves might be involved in the changes in ACP ,ALP and POD activities .

The Anesthesia Effects of Clove Oil on Koi Carp (Cy prinus carpio)

T he effects of clove oil (10 mg/L ,30 mg/L ,60 mg/L ,100 mg/L and 200 mg/L ) on anesthesia was investigated in koi carp (Cyprinus carpio) with body length of 12-15 cm and body weight of 30-40 g at water temperature of (18-20 ℃) .The anesthesia and recovery process were both divided into five stages according to the characteristics of the fish behavior .The anesthesia induction period was found to be shortened from 32 .27 min at 30 mg/L of clove oil to 4 .10 min at 200 mg/L of clove oil ,w hile the recovery time was found to be significantly prolonged from 8 .26min at 30 mg/L of clove oil to 13 .38 min at 200 mg/L of clove oil (P< 0 .01) .The recovery rate were both 100% in the fish exposed to 30 mg/L clove oil solution for 60 min and 60 mg/L clove oil solution for 30 min ;60% in the fish exposed to 100 mg/L for 20 min ;80% in the fish exposed to 200 mg/L for 10 min and 20% for 15 min .The fish exposed to air after deep anesthesia in clove oil solution had the moving time of 2 .1 min at 30 mg/L of clove oil and prolonged gradually to 25 .8 min with the increase in clove oil concentration up to 200 mg/L (P< 0 .05) .The recovery rate in the fish exposed to different concentrations of clove oil for 24 h was 0% in 35 mg/L ,60%in 30 mg/L and 100% in 10 mg/L ,15 mg/L ,20 mg/L and 25 mg/L .T he oxygen consumption rate and ammonia excretion rate of the koi carp were both found to be significantly declined with the elevated concentration of clove oil from 5 mg/L to 25 mg/L (P<0 .05) .It is suggested that 15-20 mg/L of clove oil be of optimal dose for transportation ,and 100 mg/L of clove oil be of optimal dose for handling .

Development of a Liquid Chip Technique for Simultaneous Detection of Taura Syndrome Vi rus (TSV)and Yellow Head Disease Vi rus (YHDV)

The CP2 gene of Taura syndrome virus (TSV) and N gene of yellow head disease virus(YHDV) in the GenBank were analysed by the software DNAStar 7 .0 ,and specific TSV and YHDV primers labeled with biotin were prepared and coupled with fluorescence-coded microspheres to develop liquichip technique to detect TSV and YHDV simultaneously .The aminationed probe was used for hybridization reaction to PCR products of TSV and YHDV ,and then the liquichip detection technique for detection of TSV and YHDV was established using BD FACSArray to detect fluorescence signal in the reaction system .This assay was highly sensitive to TSV ,and YHDV with a lower detection limit of 100 pg ,and was not susceptible to cross reaction with other viruses ,including white spot syndrome virus (WSSV ) ,spring viremia of carp virus (SVCV ) ,and infectious haematopoietic necrosis virus (IHNV ) .In conclusion ,the liquichip technique assay is sensitive ,and specific for TSV and YHDV detection ,and provides a novel , convenient ,and rapid approach for the detection of TSV and YHDV .

Feather Peptide Meal as Dietary Protein Sources for Eel (Anguilla marmorata)

Eel Anguilla marmorata was fed the diet containing 0% ,10% ,20% ,and 40% of feather peptide meal as fish meal replacement protein resources for feed to determine an optimum addition ratio of feather peptide meal .Results showed that the feather peptide meal was compliance with SC/T 1004-2010 and NY 5072 in good viscose-elasticity ,nutrition ,and health indicators .However ,the feather peptide meal became black in color ,glutinous ,and low content of crude fat ,calcium ,and phosphorus when it was added into the diet more .The feeding trail revealed that the optimal addition of feather peptide meal was 20% for A .marmorata grow th ,feed intake and feed conversion rate .
