The Forcast Model of Yellow Water Disease in Mud Crab Scylla serrata

It is very important to forecast yellow water disease leading to mass loss of mud crab Scylla serrata in culture ponds. According to the data Collected by monitoring station during the occurrence and spread of the disease, the survey of the correlation between the yellow water disease morbidity in the mud crab and culture environment factors revealed that there were three environment factors affecting the epidemic of the disease:rainfall, quantity of Vibrio and salinity change. With these important environment factors successfully set up one multiple linear regression forecasting model,(Y) =-0.088+0.00026TV+7.179 SC. And this forecasting model is tested by Fvalue with good results, so it can be applied to forecast the morbidity of mud crab caused by the yellow water disease.

Isolation, Determination and Bacteriostasis Test of the Bacterial Septicemia from Aemibarbus maculates

Pathogen of the Bacterial Spticemia from Aemibarbus maculates was isolated by routinely microorganism isolation procedure, and was identified with VITEK-32 as well as artificial infection test. It indicated that Aeromonas hydrophila was the major pathogen of the Septicemia. Susceptibility Test showed that the bacterium Aeromonas hydrophila was resistant to Doxycycline,and Tetracycline, Moderate sensitive to Trimeth-sulfa and highly sensitive to Enrofloxacin,Norfloxacin,Gentamicin,and Amikacin.Another bacteriostasis test in vitro showed that Enrofloxacin,Florfenicol,Thiamphenicol and Norfloxacin had strong inhibition activity to detected pathogen,and that their MIC were 0.065,4.0,8.0,and 1.0 μg/ml,respectively.

Embryonic and Larval Development of Triplophysa angeli

The embryonic and larval development was observed in Triplophysa angeli, and the morphological features and time of the development phases were described in detail under a microscope by digital camera. The fertilized adhesive eggs had a diameter of 0.87~1.05 mm and expanded to 1.24~1.41 mm upon absorption of water. The larvae hatched 119 h 44 min after fertilization at water temperature from 16.3 to 18.1 ℃, with the cumulative temperature of 2083.4 ℃·h. The biological zero degree for the fertilized eggs was found to be 4 ℃ with the effective cumulative development temperature of 1610.4 ℃·h. The embryonic development can be divided into 6 periods and subdivided into 35 developing stages. The longest period was observed in the organogenesis among the embryonic development. The newly hatched larvae had total body length of 4.58 mm, and the development of the larvae lasted nearly 28 days.

Fish Habitat Modeling in the East China Sea:A Case of Chub Mackerel

The generalized linear models(GLM)and generalized additive models(GAM)were established to evaluate the relationship between spatial distribution of the fishery resources including chub mackerel(Scomber japonicus)and the environmental factors such as the temperature, salinity, and primary productivity in the East China Sea(ECS)by the data of temperature, salinity, primary productivity and observed fishing catch in the ECS on March 1998,July 1999, October 1997 and January 2000. The impacts of temperature, salinity and primary productivity on the abundance of chub mackerel were analyzed using the GLM and the GAM. The model outputs were compared with interpolated outputs of observed patterns of distribution obtained through independent-sample T-test and error analysis. In the independent-sample T-test, P value of both GLM and GAM were over 0.05, indicating hat GLM had the advantage of expressing the trend of fish spatial distribution. In four surveys, the separate degree of the analogue results of GAM was higher than that of GLM.The results showed that the predicting results of GAM was more accurate than those of GLM. The predicting results of GAM were similar to the measured data in four surveys. It gives the rules of chub mackerel spatial distribution with the help of using GLM and GAM in some extent. GAM displays an advantage in predicting the stock assessment of chub mackerel in relating to the fish habitat to the surface temperature and salinity in the ECS, because more peak values occurred in the GAM outputs.

The Phylogenetic Relation of Sea Cucumber Acaudina molpadioidea

The phylogenetic relation of sea cucumber Acaudina molpadioidea from the coast in Zhejiang province were studied by analysis of 1312 bp from 18S rDNA and 692 bp from COImt DNA with homologous sequences of ten species within Holothuroidea downloaded from GenBank as well as the morphological characteristics. Molecular phylogenetic trees were constructed by Neighbor-Joining(NJ)and Maximum Parsimony(MP). The sea cucumber was found to be to Molpadiidae Holothuroidea, inferred from its Bio-Morphotyping. The 18S rDNA gene analysis reveals that the genetic distance between this sea cucumber and Pseudostichopus villosus is 0.017, that Acaudina molpadioidea belongs to Holothuroidea and is close to the Holothuroidea, Aspidochirota. Based on the COImt DNA gene analysis, there is close relationship between Acaudina molpadioidea and Pentamera lissoplaca, with long genetic distance of 0.223, not being the basis for the phylogenetic position.

Acute Toxicity of Chlorobenzene to Four Marine Aquatic Animals

The acute toxicity of chlorobenzene to swimming crab(Portuns trituberculatus), yellow drum(Nibea albiflora), shrimp(Exopalaemon carinicauda)and short-necked clam(Ruditapes philippinarum)was determined by low-hydrostatic method in a laboratory. It was found that 96h LC50 was 52, 15, 0.39 mg/L and 91 mg/L for the swimming crab, yellow drum, the shrimp and clam in the sea water with pH=7.9~8.2, a temperature of 24.1~24.5 ℃, and a salinity of 27.8~29.2. The acute toxicity of chlorobenzene to the four marine aquatic animals was arranged in a descend order as the following:shrimp > yellow drum > swimming crab > short-necked clam. The safety concentrations of chlorobenzene were 0.52, 0.15, 0.0039 mg/L and 0.91 mg/L for swimming crab, yellow drum, the shrimp and short-necked clam, respectively.

Effects of Dietary Protein Levels and Energy to Protein Ratios on Growth and Protease Activity in Red Swamp Crayfish Procambarus clarkii

Juvenile red swamp crayfish Procambarus clarkii was fed the diets containing different protein levels and energy to protein ratios to evaluate the effects of the dietary protein levels and energy to protein ratios on growth and protease activity. The crayfish had the best growth performance when fed the diets containing 31.86% of protein and E/P level of 35.85 kJ/g. The protease activity in hepatopancreas and stomach increased significantly with the increase in dietary protein. There were no significant effects of E/P levels on the protease activity in intestine, hepatopancreas and stomach. The maximal protease activity was observed in the crayfish fed the diets containing 31.47% of protein and E/P level of 40.42 kJ/g.

Purification and Characterization of Lectin from Gill of Bighead Carp(Aristichthys nobilis)

A fucose-binding lectin from gill of bighead carp(Aristichthys nobilis)was purified and characterized. The purification procedure consisted of extracting soluble proteins in 25 mmol/L Tris-HCl buffer(pH8.5), separating on DEAE-Sepharose FF ion exchange columns followed by gel filtration chromatography on Sephacryl S-200 HR and Superdex 200 10/300 GL. The purified lectin designated GANL showed a single protein band with an apparent molecular mass of 37 kD when submitted to SDS-polyacrylamide gel electrophoresis under reducing conditions. The native molecular mass of GANL determined by gel filtration on a Superdex 200 column was 220 kD and its carbohydrate content was estimated to be 13.4%. Therefore, GANL is a homomultimeric glycoprotein. The purified lectin only agglutinated native erythrocytes from rabbit without the addition of Ca~(2+). Its activity was not inhibited by any of the mono, disaccharides and glycoprotein tested except for fucose. Amino acid composition analysis revealed that the lectin contained high contents of Asp, Glu, Leu, Val, and Lys.GANL was stability with broad pH(5~11)and the optimum pH was 8.0~9.0. The lectin is thermostable with a temperature optima of 50 ℃.

Comparison of Embryonic Development in Gynogenetic and Diploid Barfin Flounder(Verasper moseri)

Japanese flounder(Paralichthy olivaceus)sperm was used to induce haploid of gynogenetic barfin flounder(Verasper moseri), and homologous frozen sperm was used to induce normal barfin flounder embryos to test the methods of inducing gynogenesis in barfin flounder. Observation and comparison of the embryonic developments showed that the haploid embryonic development was slower than the normal embryo before multicelled stage and it lasted 14 hours. The early blastula of haploid embryonic development was found to be slower than the normal embryos while the late gastrula, neurula stage, and formation of eyelens stage were the same as the normal embryos. The haploid embryos had slower development rate than the normal embryos did, and the haploid syndrome was observed in the haploid larvae while the gynogenetic embryonic development had the similar character to the normal embryo. Lower hatching rate and higher malformation were found in the gynogenetic and haploid larvae compared with the normal embryos.

The Pathogen of Red-head Disease in Yellow Catfish Pelteobagrus fulvidraco

The pathogenic bacterial strain HS807 was isolated from the liver of yellow catfish Pelteobagrus fulvidraco with the red-head disease. The strain HS807 was identified as Edwardsiella ictarda by morphology of the bacterial cultural performances,physiological and biochemical characteristics, the gene sequence of 16Sr RNA and artificial infection. The drug sensitivity showed that the isolated strain HS807 was highly sensitive to Cotrimoxazole, Ciprofloxacin, Cefalexin, Doxycycline, Ampicillin, Enoxacin, Rocephin, Neomycin and Furazolidone.

The Effects of Ginkgo Biloba Leaf Extract(GBE)on Cytochrome P4503A and Lysozyme Activity in Crucian Carp(Carassius auratus)

The effects of Ginkgo biloba leaf extract(GBE)on cytochrome P4503A and lysozyme activity in crucian carp(Carassius auratus)were studied by NaSH method. The results showed that GBE added to the diets significantly improved the cytochrome P4503A activity in crucian carp compared with the control group. The fish fed the diets supplemented with GBE in the three groups had significantly higher activities of cytochrome P4503A and lysozyme and higher resistance to disease at an optimal level of 25 mg/kg diet.

Purification and Biochemical Properties of Protease from White Leg Shrimp Litopenaeus vannamei

The protease was extracted and purified from heads of white leg shrimp Litopenaeus vannamei by Tris-HCl buffer solution, gel-filtertion with Sephadex G-100, and ion-exchange with DEAE-Sepharose F.F. The enzymological nature was studied with biochemical and electrophoretic techniques. The enzyme was purified to an electrophoretically homogenous state, reaching 14.3-fold purification with a yield of 31.9%. The extracted protease showed a single band in its relative molecular weight at 25 kD based on SDS-PAGE.The optimum pH was 7.0 for the enzymatic reaction and the optimum temperature was 60 ℃ for casein as the substrate This protease showed a good thermal stability below 50 ℃, and more than 60% of the activity still remained when the protease was cultured at 50 ℃ for 1 h. But the enzyme became inactivated rapidly at the temperature above 60 ℃, and only 4.7% of protease activity remained at 60 ℃ for 1 h. The protease activity was found to be inhibited by 10 mmol/L of EDTA(at a rate of 84%), and Fe~(2+)(53%), Ba~(2+)(43%)and Zn~(2+)(38%). However, 10 mmol/L of Ca~(2+) increased the enzyme activity,while 10 mmol/L of Mg~(2+) and Cu~(2+) had less effect on the protease activity. It is suggested that the protease from the shrimp be a type of metalloenzyme.

Historical Records and Current Status of Western Gray Whale in China Waters

The feeding ground of the western gray whale(Eschrichtius robustus)in summer was mainly located in the Okhotsk Sea. The gray whales usually migrate into the waters of China crossing Tatarskiy Strait and along the Korean peninsula. Thirty-one gray whales were recorded in the waters of China from 1917 to 1997. A female gray whale with a fetus was found in the coastal water of Zhejiang province in 1933. In the spring of 1953, the fishermen of Wailuo port captured two pairs of female and pup gray whales. Therefore, the coastal water area off Guangdong province was regarded as a breeding ground for the western gray whale population. Currently, the western gray whale population was facing a danger of extinction, estimated to total around 130 individuals by the survey in feeding ground-the water off Sakhalin Island and has been as "Critically Endangered" by the World Conservation Union(IUCN)in 2000. So it was of great significance to conduct investigation and research on the breeding ground of the population for the protection of the species.

Artificial Propagation of Reared Brycon cephalus

A total of 39 broodstock of 4 to 5 year old Brycon cephalus(female with body weight of 2.5~2.9 kg and body length of 36~41 cm; male with body weight of 1.5~1.6 kg and body length of 32~35 cm)was induced to spawn by two hormonal injection at an interval of 6~7 hours at a rate of HCG 300 IU +LHR-A_2 2.5 μg + DOM 2 mg/kg,HCG 3500 IU+PG 2 mg+LHR-A_2 3 μg +DOM 3 mg/kg,PG 4 mg+LHR-A_2 6 μg+DOM 2 mg/kg, HCG 700 IU+DOM 2 mg/kg, and LHR-A_2 5 μg+DOM 4 mg/kg at water temperature of 26~28 ℃ from 2007 to 2008. There were 75.2%, 75.4%, 75.5%, 60% and 60.3% of the broodstock which spawned 9~10 hours after the injection with fertilization success of 76.6%. Total of 59560 fertilized eggs and 23469 larvae(incubation period of 17~18 h) were obtained with hatching rate of 68.5%.

Cilico Cloning and Verification of eIF4A Gene from Laver Porphyra yezoensis

The silico cloning, verification and characterization of eukaryotic translation initiation factors 4 A(eIF4A)genes was conducted in cDNA of laver Porphyra yezoensis(PyeIF4A). The highly similar P. Yezoensis ESTs were obtained by Arabidopsis thaliana eIF4A amino acids sequence as a querying probe from dbEST of GenBank and the putative cDNA sequence of PyeIF4A was assembled. Furthermore, the cDNA of PyeIF4A was isolated by RT-PCR with two primers designed in accordance with the assembled sequence and was found to contain an 1278 nt of continuous complete open reading frame encoding a polypeptide(PyeIF4A)of 425 amino acids with a calculated molecular weight of 47.4 kDa. Sequence comparison of the PyeIF4A reveals high amino acid sequence identity with eIF4A from other organisms, such as mice Mus musculus, toad Xenopus laevis, rice Oryza sativa and plant Arabidopsis thaliana. The PyeIF4A contained conserved amino acids residue and sequences motifs closely related to function of eIF4A.

Sequence Comparison of Mitochondrial 12S rRNA Genes between Two Crab Species, Macrophthalmid dilatatum and M. japonicus

Partial sequences of the mitochondrial 12S rRNA gene were determined for two species crabs, Macrophthalmus dilatatum and M. Japonicus. The results showed that the 12S rRNA gene had 577 bp for M. Dilatatum and 572 bp for M. Japonicus with similar contents of A(39.0 % for M. Dilatatum;39.3 % for M. Japonicus), T(35.7 %, 36.2 %), G(16.5 %,15.8 %)and C(8.8 %,8.7 %). There were 75 different sites(except 11 deletion/insertion sites)between the two species, including 42 transition sites and 33 transversion sites. It was found that the si/sv and ratio of different sites were about 1.3 and 13.18 %, respectively. Furthermore, 331 bp homologous segments were analyzed to discuss the phylogenetic relationship between four Macrophthalmid crabs and other 23 Ocypodoid crabs in the world. The results showed that the average A+T content(76.6 %)was higher than G+C, and there were 52 variable sites and 25 parsimony-information sites in the nucleotides of the four Macrophthalmid crabs data. Neighbor-Joining method revealed that topological structure of the molecular phylogenetic tree constructed by 12S rRNA gene in all species of Macrophthalmid crabs were clustered into one distinct clade, indicating that the monophyly of Macrophthalmid crabs was well supported.

The Hybridization between Sea Urchins, Strongylocentritus nudus(♀)and Strongylocentrotus intermedius(♂)

Two species sea urchins, Strongylocentrotus nudus(♀) and S. Internidius(♂), were hybridized with the average fertilization success of about 2.5%. When the eggs of S. Nudus were fertilized with sperm of S. Intermedius, the amount of sperm selected synchronously was used at a rate of above 40 times than that in normal fertilization. Besides, the hybrid larvae had normal growth and metamorphosis at suitable water temperature, light and feeding regimes, and the period of whole metamorphosis were almost the same as their broodstocks. The hybrids had intermediate body color and thorns between their broodstocks. Total of 0.6×10~4 individuals/m~3 was obtained with test diameter from 2.59 to 2.88 mm,average of 8.8 mm during 6~7 month rearing.

The Effect of Cerium on SOD and POD Activities in Hepatopancreas of Oriental Weatherfish(Misgurnus anguillicaudatus)Stressed by Cadmium

The effects of cerium on activities of superoxide dismutase(SOD), and peroxidase(POD)in hepatopancreas of oriental weatherfish(Misgurnus anguillicaudatus)stressed by various cadmium levels. The fish were randomly divided into control group and experimental groups and exposed to solely Ce~(3+) at a rate of 0.025, 0.25 and 2.5mg/L, solely Cd~(2+) at a rate of 0.025, 0.25 and 2.5mg/L, and both Cd~(2+) and Ce~(3+) at a rate of 0.025 mg/L Cd~(2+)+0.025 Ce~(3+), 0.25 mg/L Cd~(2+)+0.25 mg/L Ce~(3+) and 2.5 mg/L Cd~(2+)+2.5 mg/L Ce~(3+).The activities of SOD, and POD were detected in the hepatopancreas at 1, 2, 3, 5, 7, and 10 days after the exposure. The SOD and POD activities were found to be significantly (P<0.05) or very significantly(P<0.01)higher in the low Ce~(3+) group than that in the control. The higher and lower SOD activities were observed in middle Ce~(3+) group in the 3d and the 5d, while the higher and lower POD activities appeared in the same group in 5d and in 7 d compared with the control(P<0.05 or P<0.01). The higher and lower SOD activities were found in high Ce~(3+) group in 2 d and from 3 d, while the higher and lower POD activities occurred in the same group in 5d and in 7d compared with the control(P<0.05 or P<0.01). The SOD and POD activities were higher in low Cd~(2+) group than those in the control(P<0.05 or P<0.01). The SOD activity in middle Cd~(2+) group was higher in 7d and lower in 10d,while the POD activity in the same group was higher all the time compared with the control(P<0.05). The SOD and POD activities in high Cd~(2+) groups were higher than those in the control in 2 d. However, the SOD activity was significantly lower in 5 d and the POD activity was lower in 3d compared with the control(P<0.05 or P<0.01). The SOD and POD activities in low and middle Cd~(2+)+Ce~(3+) groups were significantly higher, but lower in high Cd~(2+)+Ce~(3+) groups compared with the control(P<0.05 or P<0.01), indicating that low concentration cerium enhanced the activities of SOD and POD, but high concentration cerium inhibited the activities.

The Effects of Salinity on Embryonic Development of Crayfish Procambarus clarkii

Experiments were conducted to examine the effects of salinity fluctuation on embryonic development of crayfish Procambarus clarkii. The fertilized zygotes were found to hatch out under the temperature of less than 25 ℃ and a salinity of 0~8. Higher hatching rates and more larvae were observed within salinity ranging from 0 to 4. Beyond this salinity range, however, the hatching rate dramatically decreased, and incubation period prolonged. Hence, a conclusion can be drawn from these experiments that salinity from 0 to 4 was particularly suitable for the embryonic development of the crayfish with relatively high hatching rate and short incubation period.

Effects of the Interaction of Lead, Zinc and Copper Ions on Accumulation of Cadmium in Crucian Carp Carassius auratus

The effects of the interaction of heavy metals lead, zinc and copper ions on accumulation of cadmium were studies in crucian carp Carassius auratus by an ecological method. The results showed that the accumulation of cadmium in tissues of Crucian carp was dependant on the species, contents and tissue types. The accumulation of cadmium was affected not only by single metal, but also by interaction of the metals. The accumulation of cadmium was found to decrease in the gills, and to increase in the hepatopancreas. There was no significant effect of the metal mixture on the cadmium accumulation in the muscle. The lead accumulation in the tissues was in a decent order ranged as:hepatopancreas>gill>muscle.
